Human CD50 is an adhesion molecule that interacts with LFA-1 (CD18/CD11a) and provides a costimulatory signal for T cells which is thought to be essential for initiation of immune responses. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Fixed paraffin sections Flow cytometry: 5ug/ml labels 5x10e5 cultured Jurkat-4G cells Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Wash five x 10e5 cultured Jurkat-4G cells and incubate 45 minutes on ice with 80ul of C2405-06B at a concentration of 5ug/ml. 2. Wash cells twice and incubated with Streptavidin/R-PE. 3. Wash cells three times, fixed and analyzed using FACS. 4. Cells stained positive with a mean shift of 2.64 log10 fluorescent units when compared to a Mouse IgG2b negative control at a similar concentration. 5. Binding was 99% blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml of C2405-06A CD50. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
