Human CD54 (ICAM-1) mediates cell adhesion by binding to the integrins CD11a/CD18 (LFA-1) and to CD11b/CD18 (Mac-1). CD54 expression on resting peripheral blood leukocytes in weak but is upregulated on activated T and B lymphocytes and on monocytes. Applications: Suitable for use in FLISA, Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 cultured human Daudi cells were washed and preincubated ~5 minutes with 20ul of 0.25mg/ml human IgG (to block non specific binding) after which they were incubated 45 min on ice with 80ul of C2409-10D at a dilution of 1:50. 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.78 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 4. Binding was 99% blocked when preincubated 10 min with 20ul of C2409-10A at a concentration of 0.5mg/ml. Storage and Stability: May be stored at 4C before opening. DO NOT FREEZE! Stable at 4C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
