Human CD59 is a GPI anchored molecule that protects cells from complement-mediated lysis (1). Studies are progressing to determine the regulatory mechanism for CD59 expression (2). Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 (~80ul/test) Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 cultured HPB-MLT cells were washed and preincubated ~5 minutes with 20ul of 250mg/ml human IgG (to block nonspecific binding). 2. After which they were incubated 45 minutes on ice with 80ul of C2413-21R at a dilution of 1:50. 3. Cells were washed three times, fixed and analyzed using a BD FACS Calibur. 4. Cells stained positive with a mean shift of 2.81 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 5. Binding was 92% blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml C2413-21N. Storage and Stability: May be stored at 4C before opening. DO NOT FREEZE! Stable at 4C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
