Quantity: 30 assays per kit Quality Control Testing: This kit was used to precipitate cdc42-GTP from 1mg of a 3T3 cell lysate that had been loaded in vitro with GTP gamma-S. Previous lots precipitated cdc42-GTP from HL-60 and HeLa cell lysates that had been loaded in vitro with GTP gamma-S, as well as from 1mg of lysate from PMA-stimulated HL-60 cells that had been differentiated to neutrophillike cells by treatment for 5 days with 1.4% DMSO. The precipitated cdc42-GTP was detected by Western Blot using anti-cdc42 (1ug/ml). Kit Components: C2562-80A: PAK-1 PBD (Agarose), 1x300ul. GST-fusion-protein corresponding to the p21-binding domain (PBD, residues 67-150) of human PAK-1 conjugated to agarose. C2562-80B: cdc42 Mab, 1x50ug. Purified monoclonal mouse IgG1. C2562-80C: Lysis/Wash Buffer (5X), 2x18ml. C2562-80D: GTP gamma-S (100X), 1x50ul. Non-hydrolyzable analog of GTP (Guanosine 5-O-(3-thiotriphosphate). C2562-80E: GDP (100X), 1x50ul. GDP (Guanosine 5- Diphosphate) for in vitro labeling of G-proteins in the inactive form. Storage and Stability: Kit compononents are stable for 12 months after receipt at -20C. For maximum recovery of product, centrifuge the original vials prior to removing the cap.
