MaxLight(TM)550 is a new Yellow-Green photostable dye conjugate comparable to Alexa Fluor(TM)546, 555, DyLight(TM)549 , Cy3(TM), TRITC and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (550nm), Emission (575nm), Extinction Coefficient 150,000. Type IV collagen is a major component of the basement membrane and it associates with laminin, entactin and heparin sulfate proteoglycans to form sheet-like structures that separates epithelium from connective tissue. It is a long triple helix containing several interruptions, which is characterized by a specific carboxylterminal, non-collagenous NC1 domain. It is composed of six genetically different alpha (IV) chains, alpha1 (IV) to alpha 6(IV) and their genes are paired on three different chromosomes in a head-to-head arrangement. The alpha 1 gene and the alpha 2 gene are localized on chromosome 13q34, the alpha 3 gene and the alpha 4 gene are on chromosome 2q36, and the alpha 5 gene and the alpha 6gene are on chromosome Xq22. Type IV collagen can be used as a thin coating on tissue culture surfaces to promote cell attachment and proliferation and to study its effects on cell behavior. Applications: Suitable for use in Immunofluorescence, Immunohistochemistry and FLISA. Other applications have not been tested. Recommended Dilution: Immunofluorescence: 1:40-1:160 Immunohistochemistry (paraffin): 1:250-1:1000 (Reccommended Immunohistochemistry fixatives: AFA, buffered formalin (5-10%) or Bouins) FLISA: 1:100-1:400 Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1903-30U: IgG, H&L, X-adsorbed (HRP) ShxRb Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)550 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
