Collagenases degrade native helical collagen fibrils. The substrate, collagen, is the major fibrous component of animal extracellular connective tissue: skin, tendon, blood vessels, bone, etc. Suitable for use in cell dissociation procedures. Collagenase is prepared to contain lower tryptic activity levels to limit damage to membrane proteins and receptors but with normal to above normal collagenase activity. Suitable for use in pancreatic islet primary isolation and other applications where receptor integrity is crucial. Cofactors: Zinc, Calcium Collagenase nomenclature: Type 1 Containing average amounts of assayed activities (collagenase, caseinase, clostripain and tryptic activities). It is generally recommended for fat, adrenal, and liver cells. Type 2 Containing greater clostripain activity. It is generally used for heart, bone, muscle, thyroid, cartilage, and liver cells. Type 3 Containing low proteolytic activity. It is usually used for mammary and fetal cells. Type 4 Containing low tryptic activity. It is commonly used for islets and other applications where receptor integrity is crucial. Source: Clostridium histolyticum Form: Supplied as a dialyzed, lyophilized powder. Activity: 100units/mg dry weight. Unit Definition: 1 unit releases 1 µmole of L-leucine equivalents from collagen in 5 hours at 37C, pH 7.5. Quality Control: SDS-PAGE Storage and Stability: Collagenase is stable for 1-2 years at 2-8C. Storage at -20C (desiccated) is also acceptable.
