Human Cot/Tpl2 (GenBank Accession No. NM_005204), (aa30-397), with N-terminal GST-HIS6-tag, expressed in a Baculovirus infected Sf9 cell expression system. Specific Activity: ~40pmol/min/ug Assay Conditions: 100ul of 240nm biotinylated MEK1 peptide was coated onto a neutravidin plate for 1 hour at 37C. After overnight blocking with TBS superblock, the reaction was done with 10uM ATP for 60 minutes at RT, and phospho-MEK1 was detected using an anti-phospho-MEK1 antibody. Specific activity was determined relatively by comparing with a standard kinase under the same condition. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for 6 months.
