Biotin-labeled Nucleotides are used for non-fluorescent labeling of nucleic acids in purification and detection applications. Biotin-11-dATP is enzymatically incorporated into DNA/cDNA as substitute for its natural counterpart dATP. The resulting Biotin-labeled DNA/cDNA probes are subsequently detected using streptavidin conjugated with horseradish peroxidase (HRP), alkaline phosphatase (AP), a fluorescent dye or agarose/magnetic beads. Optimal substrate properties and thus labeling efficiency as well as an efficient detection of the Biotin moiety is ensured by a 11-atom linker attached to the 7-Deaza position of adenine. Spectroscopic Properties: lambdamax 280nm, epsilon 12,700 Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
