Control Peptide for E2291-52 (antiserum) and E2291-53 (affinity purified) A 17 amino acid peptide sequence near the amino terminus of human ERAB was used to generate anti-ERAB. The peptide was coupled with KLH. An intracellular protein termed ERAB ( Endoplasmic reticulum associated Ab binding protein on chromosome X) has been cloned and linked with Ab neurotoxicity (1). ERAB has structural homology with short-chain alcohol dehydrogenases (hydroxysteroid dehydrogenases and acetyl-CoA reductases). Interestingly, ERAB localizes to endoplasmic reticulum despite the absence of a classical transmembrane sequence. However, in the presence of Ab, ERAB- Ab complex translates to the inner plasma membrane. Alzheimer patients have strong ERAB activity. Normal brains have virtually no ERAB. In addition, Ab neurotoxicity can be reduced by blocking ERAB and increased by its overexpression. Human ERAB is an intracellular (endoplasmic reticulum), 264aa (Mouse ERAB 261aa), single polypeptide of approx. 27kD. It appears to ubiquitously expressed in normal brain and other tissue (1). Although, it is expressed at a higher levels in brains from Alzheimer patients.
