Control Peptide for E2291-52A (antiserum) and E2291-53A (affinity purified antibody). An intracellular protein termed ERAB (262aa, Endoplasmic reticulum associated Ab binding protein on chromosome X) has been cloned and linked with Ab neurotoxicity. ERAB has structural homology with short-chain alcohol dehydrogenases (hydroxysteroid dehydrogenases and acetyl-CoA reductases). Interestingly, ERAB localizes to endoplasmic reticulum despite the absence of a classical transmembrane sequence. However, in the presence of Ab, ERAB-Ab complex translates to the inner plasma membrane. Alzheimer patients have strong ERAB activity. Normal brains have virtually no ERAB. In addition, Ab neurotoxicity can be reduced by blocking ERAB and increased by its overexpression. Applications: Suitable for use in ELISA and Antibody Blocking. Not suitable for use in Western Blot due to low molecular weight. Other applications not tested. Recommended Dilution: Antibody Blocking: 5-10ug per 1ui E2291-52A (antiserum) or per 1ug E2291-53A (affinity purified antibody). ELISA: Coat 50-100ng of control peptide/well. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
