Beta-1,4-Galactosyltransferase 1 (B4GalT1) is one of seven beta1, 4 galactosyltransferases that transfer galactose in a beta1,4 linkage to acceptor sugars including Gl cNAc, and Gl c, and Xyl. By sequence similarity, the B4GalTs form four groups: B4GalT1 and B4GalT2, B4GalT3 and B4GalT4, B4GalT5 and B4GalT6 and B4GalT7. B4GalT1 is unique among the seven enzymes because it can be expressed either as membrane associated form or secreted form. The secreted form is restricted to lactating mammary tissues where the enzyme forms a heterodimer with alpha-l actalbumin to catalyze the synthesis of lactose. The membrane form can reside either in the Golgi apparatus, where it adds galactose to N-acetylglucosamine residues, or on cell surface, where it functions as a recognition molecule during a variety of cell to cell and cell to matrix interactions, by binding to specific oligosaccharide ligands on opposing cells or in the extracellular matrix. The two enzymatic forms result from alternate transcription initiation sites and posttranslational processing. Defects in B4GalT1 are the cause of congenital disorder of glycosylation type 2D(CDG2D). Recombinant corresponding to Gly44-Ser398 from human B4GalT1 with an N-terminal 6 His tag expressed in NSO cells. N-terminal Sequence Analysis: His Predicted Molecular Mass: 40kD SDS-PAGE: 45 55kD Activity: Measured by its ability to transfer galactose from UDP Galactose to N-Acetyl-alpha-D-glucosamine. The specific activity is >2000pmol/min/ug, as measured under the described conditions. See protocol. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
