Placental growth hormone, or growth hormone variant (GH-V), is a member of the growth hormone gene family. Its physiologic role during pregnancy remains undefined. Although extensive work has been done characterizing the signaling pathways activated by hGH, the signaling pathways which are activated by GH-V have not been characterized. Human placental GH (hPGH) replaces pituitary GH during pregnancy. hPGH is correlated to serum IGF-I in normal pregnancies and in pregnancies complicated by fetoplacental disorders Recombinant Placental Human GH-20K is a single, non-glycosylated, polypeptide chain containing 177 amino acids and having a molecular mass of 20498 Dalton. Predicted pI=8.20. PGH-20K-pl is devoid of lactogenic (prolactin receptor mediated) activity characteristic to pituitary GHs. Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Phe-Pro-Thr-Ile. Dimers and Aggregates: 1% as determined by silver-stained SDS-PAGE gel analysis. Biological Activity: Placental Growth Hormone-20K is fully biologically active when compared to World Health Organization (WHO) reference standard. Storage and Stability: Lyophilized powder may be stored at 4C for short-term only. Reconstitute to nominal volume by adding sterile 0.4% NaHCO3 and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
