The protein encoded by this gene is a protein-tyrosine kinase that is predominantly expressed in hemopoietic cell types. The encoded protein may help couple the Fc receptor to the activation of the respiratory burst. In addition, it may play a role in neutrophil migration and in the degranulation of neutrophils. Alternate translation initiation site usage, including a non-AUG (CUG) codon, results in the production of two different isoforms, that have different subcellular localization. N-terminal His6-tagged, recombinant, human Hck, aa230-497, expressed by baculovirus in Sf21 insect cells. Specific Activity: As reported (typically >10,000U/mg). One unit of Hck activity is defined as 1nmol phosphate incorporated into 250uM Src substrate peptide (KVEKIGEGTYGVVYK) per minute at 30C with a final ATP concentration of 100uM. Quality Control Testing: MS Tryptic Fingerprint: Confirmed identity as Hck with 64% amino acid coverage of the translated native sequence Hck Kinase Assay: 1-20ng phosphorylated 250uM Src substrate peptide. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
