HIF-2-alpha/EPAS1 (hypoxia-inducible transcription factor 2-alpha/enodothelial PAS domain 1) is stabilized in hypoxic tissue and, similarly to HIF-1-alpha, complexes with Aryl hydrocarbon receptor nuclear translocator (ARNT). Both the HIF-1 and HIF-2 complexes bind hypoxia-response elements (HREs) in the promoters of many genes involved in adapting to an environment of insufficient oxygen or hypoxia. HIF-1 and HIF-2 do not appear completely redundant, although specific functions are only beginning to be elucidated. Hypoxic tissue environments occur in vascular and pulmonary diseases as well as cancer, which illustrates the potentially broad impact of gene regulation by both HIF-1-alpha and HIF-2-alpha. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using 786-O human renal cell adenocarcinoma cell line (HIF-2 alpha positive), Caki-2 human clear cell carcinoma epithelial cell line (HIF-2alpha negative), and HeLa human cervical epithelial carcinoma cell line (HIF-2-alpha negative) Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
