DNA sequences encoding the extracellular domain of human Integrin alpha M (Phe17-Asn1105, Accession P11215) (Corbi, A.L., et al., 1988, J. Biol. Chem. 263:12403) with an acidic tail at the C-terminus and the extracellular domain of human Integrin b2 (Gln23-Asn700, Accession P05107) (Kishimoto, T. et al., 1987, Cell 48:681) with a basic tail at the C-terminus were cloned downstream of the human CD33 signal peptides and co-expressed in Chinese hamster ovary (CHO) cells. Specifictity: The 1088aa human aM/CD11b ECD shares 73-76% aa sequence identity with mouse, rat, bovine and canine aM while the 678aa human b2/CD18 ECD shares 81-83% aa sequence identity with mouse, rat, bovine, canine, goat, sheep, and porcine b2. Molecular Mass: Based on N-terminal sequencing, the recombinant human Integrin aM subunit starts at Phe 17 and has a calculated molecular mass of ~124.7kD. The N-terminal sequence of the b2 subunit was blocked, suggesting that the b2 subunit may start at the predicted Gln 23 and has a calculated molecular mass of a~79kD. As a result of glycosylation, the recombinant proteins migrate as ~165 and 100kD protein bands in SDS-PAGE under reducing conditions. Activity: Measured by the ability of the immobilized protein to support the adhesion of CHO cells transfected with human ICAM-1. The ED50 for this effect is 0.25-1.5ug/ml. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
