The specificity of mAb IPO-38 was demonstrated in different human and murine lymphoid and nonlymphoid cell lines. It does not react with mononuclear cells and granulocytes of healthy donors but does react with nuclei of blood cells of patients with different forms of leukemia and with cells from lymph nodes of patients with Hodgkins disease and non-hodgkins lymphoma. Also a positive reaction in nuclei of breast and colorectal tumor cells was demonstrated. IPO-38 may be a good marker of cellular proliferation during monitoring of tumor progression. Applications: Suitable for use in ELISA, Immunoprecipitation, Western Blot and Immunohistochemistry frozen and formalin/paraffin). The antibody is reported to recognize a nuclear antigen that is present in the cytoplasm and nuclei of proliferating cells paraformaldehyde fixed, Triton X-100 permeabilized). An increase of 400% is observed in cells in mitosis (determined by analysis of K562 cells synchronized with colcemide). IPO-38 does not block the binding of the Ki-67 antibody. Recommended Dilutions: ELISA: 1:1000. Immunohistochemistry:1:50 to 1:100 Western Blot: 1:50 to 1:100 Optimal dilution determined by the researcher. Positive Control: Tonsilar tissue Hybridoma: BALB/c mouse splenocytes and mouse P3-X63-Ag5.653A myeloma cells. Storage and Stability: May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
