The mitogen-activated protein kinase (MAP) kinase family includes ERK, JNK and p38 kinase. The pathway, regulated by (MAP) kinase family, involves a broad array of cellular responses that include survival, cell proliferation, and apoptosis. The JNK signal transduction pathway is activated in response to environmental stress and by engagement of several classes of cell surface receptors including cytokine receptors, serpentine receptors and receptor tyrosine kinases. Although JNK is located in both the cytoplasm and the nucleus of quiescent cells, activation of JNK is associated with accumulation of JNK in the nucleus. In the NGF (nerve growth factor)-differentiated PC-12 cells, activation of the ERK signaling pathway promotes cell survival but stimulation of the JNK and p38 signaling pathways contributes to cell death. Applications: Suitable for use in Immunofluorescence and Western Blot. Other applications have not been tested. Recommended Dilution: Immunofluorescence: 5-10ug/ml Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
