Ki67 antigen is the prototypic cell cycle related nuclear protein, expressed by proliferating cells in all phases of the active cell cycle (G1, S, G2 and M phase). It is absent in resting (G0) cells. Ki67 antibodies are useful in establishing the cell growing fraction in neoplasms. In neoplastic tissues the prognostic value is comparable to the tritiated thymidine-labelling index. The correlation between low Ki67 index and histologically low-grade tumors is strong. Ki67 is routinely used as a neuronal marker of cell cycling and proliferation. Applications: Suitable for use in Immunohistochemistry, Immunocytochemistry and Immunofluorescence. Other applications not tested. Recommended Dilutions: Immunohistochemistry (Frozen/Paraffin): 1:50-1:300 Formalin-fixed paraffin-embedded tissue sections require high temperature antigen unmasking with 10mM citrate buffer, pH 6.0 prior to immunostaining. Will not work without optimal antigen retrieval. An incubation period of 30 minutes at RT and the use DAB to stain the protein (immunofluorescence may give problems as the protein is nuclear) is suggested. Immunocytochemistry: 1:50 Immunofluorescence: 1:20 Optimal dilutions to be determined by the researcher. Positive Control: Tonsil Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
