Methionine Aminopeptidase 1D (MAP1D) is a member of the Methionine Aminopeptidase family (MetAPs) (1). MetAPs catalyze the removal of the initiator methionine residue from nascent peptides (2) and are essential for cell growth (3). MAP1D is localized in mitochondria (4) and is overexpressed in colon cancer cell lines and colon tumors (1). The N-terminal leader peptide (residue 1-43) of MAP1D has been shown to be responsible for targeting to mitochondria (4). Source: Recombinant human Methionine Aminopeptidase 1D (rhMAP1D, residues 44-335, Accession NP_954697) was expressed with a C-terminal 10X His tag in an insect cell line, Sf 21. Definition: Measure by its ability to remove methionine from a fluorogenic peptide substrate Met-Gly-Pro-AMC in a coupled assay. Specific Activity: 30 pmol/min/ug. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: Predicts 34kD and migrates as a 38kD band in SDS-PAGE under reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
