Mucins are glycoproteins abundant with O-linked oligosaccharides secreted by epithelial mucous membranes. Their protective function is due to their high viscosity. Bovine submaxillary mucin has a molecular weight of 4-40x10e5 Daltons. The molecule consists of major and minor components with a protein moiety (36.6% of the molecule) and a carbohydrate moiety (56.7% of the molecule). Mucins are heavily glycosylated glycoproteins, which constitute the major components of mucus which cover the surface of epithelial tissues. Nine distinct epithelial mucin genes (Muc-1, 2, 3, 4, 5AC, 5B, 6,7 and 8) have been identified. Various immunohistochemical and in situ hybridization studies have shown that these mucins are differentially expressed in epithelia with cell-type specificity. Muc-5AC and Muc-6 glycoproteins are the first two mucins found in superficial epithelium and Muc-6 glycoprotein is found in the deep glands. Applications: Suitable as a substrate for neuraminidase assays. Source: Bovine submaxillary gland Purity: Mucin is prepared by the method of Nisizawa and Pigman [The composition and Properties of the Mucin Clot from Cattle Submaxillary Glands, Arch. Oral Biol., 1:16 (1959)]. Bovine sub-maxillary glands are blended in a phosphate buffer, the material is filtered followed by a pH adjustment, then centrifuged and the ppt is resuspended, a final pH adjustment is made and the material is put in dialysis prior to lyophilization.
