PACSINs, also known as syndapins, are a protein family consisting of three members that have been implicated in synaptic vesicular membrane trafficking and regulation of dynamin-mediated endocytotic processes (Cuajungco, 2006). It is presumed that all isoforms bind Dynamin, Synaptojanin 1 and N-WASP through their Src homology 3 domains. While PACSIN1 appears to be restricted to neural tissues, two PACSINs are more widespread with PACSIN2 being ubiquitously expressed and PACSIN3 present mainly in skeletal muscle, heart and lung (Modregger, 2001). PACSIN1 has been implicated as playing a central role in synaptic vesicle recycling (Kessels, 2004). Expected molecular weight is 51kD. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:100-1:500 Immunohistochemistry: Paraffin sections Optimal dilutions to be determined by the researcher. Positive Control: Mouse brain lysate Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
