Human PI3K (p120 gamma) (GenBank Accession No. AF327656), full length with N-terminal His tag, expressed in an Baculovirus infected Sf9 cell expression system (MW:~131kD). Specific Activity: Lot-dependent. Sample Value: ~1200 pmol/min/ug. Reaction: A 50ul kinase reaction is conducted in a buffer containing 40mM Tris-HCl (pH 7.5), 20mM MgCl2, 100uM ATP, 200uM L-alpha-phosphatidylinositol/ L-alpha-phosphatidylserine (PI/PS), and 20-200ng enzyme for 15 min at 30C. ATP reduction is detected using ADP-Glo(TM) Kinase Assay Platform (Promega Corporation). Application: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Storage and Stability: Store at -70C. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
