Plasminogen Activator Inhibitor 1 (PAI-1) is a glycoprotein found in plasma that is implicated in the processes of angiogenesis, hemostasis, tumor metastasis, fibrionlysis, ovulation, embryonic development and inflammation (1). The elevation of PAI-1 has shown to correlate with the prognosis of many cancers including breast cancer (2,3,4). PAI-1 is activated by the glycoprotein vitronectin. Vitronectin binds to PAI-1 and modulates fibrinolysis and cell migration (5). This binding has proven to be an important inhibitor of the activated form of protein C (APC) (6). The Mouse PAI-1 have a single mutation, I91L, to create an enhanced half-life as compared to the wild type form. Absorbance (280nm E0.1% = 1.0) Kinetic Data: Second order rate constants for inhibition of: uPA = 6.3 X 10e6 M -1s -1 tPA = 5.7 X 10e6 M -1s -1 Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Half-life at 37C and pH 7.4 is ~18 hours.
