PDGF-BB (Platelet derived growth factor B chain), a disulphide-linked beta-chain homodimer and member of the PDGF family, released from platelet alpha-granules following trauma/platelet activation. Naturally occurring active forms of PDGF exist as dimers of two alpha chains (AA), two beta chains (BB), or a combination of both chains (AB), all of which signal through the high-affinity PDGF-Ralpha receptor, and the latter two of which can also signal through the PDGF-Rbeta receptor. PDGFs are potent mitogens for a variety of cells including those of the connective tissue, bone and cartilage, blood and smooth muscle cells, and are involved in several biological processes including chemotaxis, hyperplasia and embryonic neurogenesis. The expression of PDGF-BB is associated with autocrine growth stimulation and the formation of various tumor types, with several studies focusing on dermatofibrosarcoma protuberans (DP), an infiltrating skin tumor arising from a translocation occurring between chromosome 22 (the location of the PDGFB gene) and chromosome 17 (the location of the COL1A1 gene), which results in the generation of the COL1A1-PDGFB chimeric protein. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilutions: ELISA (sandwich): 0.5-2ug/ml (using 100ul/well antibody. In conjunction with P4258-18 as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hPDGF-BB. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hPDGF-BB is 1.5-3ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
