| This kit contains three fluorochrome conjugated antibodies that can be used for single-step immunocytochemical staining of human pluripotent stem cells (including embryonic and induced pluripotent stem cells). Each antibody is supplied as 12.5ug in 125ul of PBS containing 0.1% sodium azide. Kit Components: Anti-human SOX2 NL557-Conjugated Goat IgG Anti-human Oct-3/4 NL637-Conjugated Goat IgG Anti-human Nanog NL493-Conjugated Goat IgG Intended Use: This product is designed for the immunocytochemical analysis of human pluripotent stem cells using three fluorochrome-conjugated antibodies. Immunocytochemistry Validation: These antibodies have been tested for immunocytochemistry staining using BG01V cells grown on irradiated mouse embryonic fibroblasts. Cells were fixed in PBS containing 4% paraformaldehyde and blocked with PBS containing 10% normal donkey serum, 0.3% Triton X-100, and 1% BSA. After blocking, cells were incubated for 3 hours at RT in the dark with all 3 conjugated antibodies at a final concentration of 10ug/ml each (1:10 dilution) in blocking buffer. Between each step, cells were washed with PBS containing BSA. When using a staining volume of 50ul, this kit contains 25 tests. Fluorochrome Absorption Maximum (nm) Emission Maximum (nm) NL557 557 574 NL637 637 658 NL493 493 514 Typical Data: BG01V cells grown on irradiated mouse embryonic fibroblasts were simultaneously stained with (A) NL637- conjugated Oct-3/4, (B) NL493-conjugated Nanog, and (C) NL557-conjugated SOX2. All nuclei were stained with DAPI (shown in panel D). Storage and Stability: Store at 2-8C in the dark. Use within 6 months of receipt. BG01V cells are licensed from Novocell, Inc.
