S100 belongs to the family of calcium binding proteins such as calmodulin and troponin C. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains. Antibody to S100 stains Schwannomas, ependymomas, astrogliomas, almost all benign and malignant melanomas and their metastases. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes. S100 is a nervous tissue specific soluble protein made in glial cells. Present in sweat glands chongrocytes and myoepithelial cells. May be found as cytoplasmic, membranous, or mixed throughout the areas of tumor. Usually cytoplasmic when present in normal tissues. S100 Protein derived from brain tissue is an acidic calcium-binding protein with the molecular weight of ~21kD. In human brain tissue S100 protein is mainly presented by two isoforms, beta-beta homodimer (S100b) and alpha-beta heterodimer (S100a). Applications: Suitable for use as a standard in immunoassay, as an immunogen for antiserum production and tracer for iodination. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. (Add fresh 2-mercaptoethanol to a concentration of 5mM after prolonged storage.) Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
