The AMP-activated protein kinase (AMPK) cascades appear to be sensors for cellular energy status. The members of the AMP-activated/SNF-1-related subfamily of protein kinases are thought to be pivotal components of these highly conserved protein kinase cascades, which are in most eukaryotic cells. The SAMS peptide is based on the 13-residue sequence around Ser79, which is the unique site for the AMP-activated protein kinase on rat acetyl-CoA carboxylase. The serine at residue 77, which corresponds to the cyclic-AMP-dependent protein kinase site in the natural sequence of rat Acetyl-CoA Carboxylase (ACC), was replaced by alanine at residue 5 of the synthetic sequence. Two arginine residues were added at the C-terminus to increase binding of the peptide to phosphocellulose paper. Amino Acid Sequence: HMRSAMSGLHLVKRR Applications: Suitable for use in Protein Phosphorylation Assays. Other applications not tested. Recommended Dilutions: Protein Phosphorylation Assays: Use 10-100mU/assay point. Optimal dilutions to be determined by researcher. Peptide Phosphorylation Assay: Tested by using 20mU of AMPK activated enzyme to phosphorylate 100uM SAMS substrate peptide in the presence and absence of 300uM AMP. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
