Produktbeschreibung

SHP/SHPTP are tyrosine phosphatases which contain SH2 domains. SHP 1/SHPTP is expressed primarily in hematopoietic cells, and binds specific phosphotyrosine residues, often in ITIM motifs. SHP 1 is a down-regulator of receptor signaling and inhibits activation of macrophages, B-cells, T-cells, and mast cells. Motheaten mice harbor a mutation in SHP 1, which leads to progressive inflammation. SHP 2 has a broader expression pattern, and appears to promote cell proliferation and differentiation. Additionally, SHP 2 regulates cell spreading and migration in a manner similar to FAK, suggesting that the two act together to dynamically remodel focal adhesions. Recombinant protein corresponding to aa224-529 in the catalytic domain of human SHP 2 with N-Terminal GST-Tag, expressed in an E. coli expression system. GenBank Accession: NM_002834 Specific Activity: ~3U/ug. Unit Definition: One unit will hydrolyze 1nmol p-nitrophenyl phosphate per minute at pH 7.4 and 30C Assay Buffer: 50mM HEPES, pH 7.4, 2mM EDTA, 3mM DTT, 100mM sodium chloride, 50mM pNPP. Incubate for 20 minutes at 30C. Absorbance is measured at 405nm. Applications: Sutiable for use in Enzyme Kinetics, Screening inhibitors and Selectivity Profiling. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
