Human Sirtuin 3 (GenBank Accession No. NM_012239), amino acids 102- 399 with N-terminal GST tag, expressed in E. coli expression system. Specific Activity: 6 U/ug. One U =1 pmol/min Assay Condition: 25mM Tris/Cl, pH 8.0, 137mM NaCl, 2.7mM KCl, 1mM MgCl2 and 0.1mg/ml BSA, 30uM BPS HDAC substrate, 200uM NAD+ and 20ng/ul Sirtuin 3. Incubation Condition: 30 min at 30C Application: Useful for the study of enzyme kinetics, screening inhibitors and selectivity profiling. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
