SOC broth (Super Optimal Broth with Catabolite repression) is a nutrient-rich liquid growth medium used in molecular biology, especially for recovering E. coli bacteria after genetic transformation (inserting DNA), boosting the survival and expression of antibiotic resistance genes before plating on selective media, containing ingredients like tryptone, yeast extract, salts, and glucose for quick cell repair and growth. Applications: 1. Recovery Medium: Helps E. coli recover from the stress of transformation (heat shock or electroporation). 2. Enhances Efficiency: Increases the number of successful transformants (colonies with new DNA). 3. Allows Gene Expression: Provides nutrients for cells to express antibiotic resistance genes before they encounter antibiotics on agar plates. Usage in Cloning: 1. Transform Cells: Introduce DNA into E. coli. 2. Add SOC: Mix the transformed cells with SOC broth and incubate briefly (e.g., 30-60 mins). 3. Plate: Spread on agar plates containing the appropriate antibiotic to select for successfully transformed cells.
