N-terminus, His-tagged fusion protein corresponding to full-length human Syk as confirmed by Ms tryptic fingerprinting and protein sequencing, expressed in Sf21. Applications: Suitable for use in MS Tryptic Fingerprint and Kinase Assay. Other applications not tested. Recommended Dilution: MS Tryptic Fingerprint: Confirmed identity as Syk with translated sequence. Kinase Assay: 20-55ng phosphorylated 0.1mg/ml poly(Glu, Tyr) (4:1). Optimal dilutions to be determined by the researcher. Specific Activity: 195U/mg, One unit of Syk activity is defined as 1nmol phosphate incorporated into 0.1mg/ml poly(Glu, Tyr) (4:1) per minute at 30C with a final ATP concentration of 100uM. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Handling Recommendations: Rapidly thaw the vial under cold water and immediately place on ice. Aliquot unused material into pre-chilled microcentrifuge tubes and immediately snap-freeze the vials in liquid nitrogen prior to re-storage at -70C.
