TPMT, thiopurine S-methyltransferase, is a cytosolic enzyme that metabolizes thiopurine drugs via S-adenosyl-L-methionine as the S-methyl donor and S-adenosyl-L-homocysteine as a byproduct. Thiopurine drugs such as 6-mercaptopurine and azathioprine are used as chemotherapeutic agents. TPMT activity exhibits autosomal codominant genetic polymorphism, and patients inheriting TPMT-deficiency are at high risk of potentially fatal hematopoietic toxicity. Recombinant human TPMT was expressed in E.coli and purified by using conventional chromatography techniques. This product is recommended for use in applications that do not require a catalytically active form of the protein. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
