The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. The TRL family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. Ten human homologs of TLRs (TLR1-10) have been described. TLR8 gene contains three exons, two of which have coding function. TLR8 cDNA codes for a protein of approximate molecular weight of 120kD. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry (Intracellular): 0.5-2ug labels 1x10e6 cells. Ramos cells can be used as a positive control. Western Blot: A band of ~120kD was detected with transfected cell lysates. Immunohistochemistry: Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Positive Control: Western Blot: Transfered cell lysate Flow Cytometry: Raw or Raw cells Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
