Immobilized anhydrotrypsin selectively adsorbs peptides with Arg, Lys or AECys (S-aminoethyl cysteine) residues at the C- terminus under weak acidic conditions (1). Strongly acidic conditions are used to elute the bound peptides. Peptides with these amino acids at the N-terminus or internally, as well as free amino acids are not bound. Serine protease inhibitors such as ecotin and PAI-1 may also be purified on this resin (2). May be used repeatedly. For long-term storage, aliquot and store at 4C. Do not freeze. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
