Troponin, working in conjunction with tropomyosin, functions as a molecular switch, regulating muscle contraction in response to changes in the intracellular Ca2+ concentration. Troponin consists of three subunits, the Ca2+-binding subunit troponin C (TnC), the tropomyosin-binding subunit troponin T (TnT), and the inhibitory subunit troponin I (TnI). Troponin I binds to actin in thin myofilaments to hold the actin-tropomyosin complex in place, because of which myosin cannot bind to actin in relaxed muscle. When Calcium binds to the Troponin C it causes certain confomational changes which leads to dislocation of troponin I. The letter ''I is given due to its inhibitory character. The tissue specific subtypes are: Slow-twitch skeletal muscle isoform troponin I, TNNI1 (1q31.3, 191042) Fast-twitch skeletal muscle isoform troponin I, TNNI2 (11p15.5, 191043) Cardiac troponin I, TNNI3 (19q13.4, 191044) Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
