Soybean trypsin inhibitor (SBTI) is a reversible competitive inhibitor of trypsin and other trypsin-like proteases such as chymotrypsin, plasmin and plasma kallikrein. This presentation of SBTI binds free trypsin tightly with high pH (4.5-8.0) and allows for a simple gradient to low pH (~2.5-3.0) for elution.1 This presentation immobilized SBTI is on an agarose resin via coupling to primary amines. This creates an affinity resin with specificity for a number of serine proteases. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
