Soybean trypsin inhibitor (SBTI) is a reversible competitive inhibitor of trypsin and other trypsin-like proteases such as chymotrypsin, plasmin and plasma kallikrein. This presentation of SBTI binds free trypsin tightly with high pH (4.5-8.0) and allows for a simple gradient to low pH (~2.5-3.0) for elution.1 This presentation immobilized SBTI is on an agarose resin via coupling to primary amines. This creates an affinity resin with specificity for a number of serine proteases. Buffer: 0.1 M Tris, 0.05 M NaCl, 0.1% Sodium Azide, pH 7.5 Isoelectric Point: 4.5 (1,2) Molecular Weight: 22 kDa (1,2) Detail as follows: -Resin Matix: Affi-Gel 10 -Attachment: Amino -Ligand Density: 1-2 mg/mL SBTI -Binding Capacity: 1 mL resin binds > 2 mg plasmin Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 6 months after receipt at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
