MaxLight(TM) 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor(TM)647, DyLight(TM)649, Cy5(TM) and offers better labeling efficiency, brighter imaging and increased immunodetection. The tubulin protein is a major target of drug molecules, and consequently, tubulin inhibitors have attracted great attention as antimitotic antitumor agents for chemotherapeutic use. Stable microtubules (MTs) are known to contain acetylated alpha-tubulin. Acetylated tubulin is found in the dense networks and bundles of microtubules in the sensory and supporting cells. MTs containing acetylated alpha-tubulin in Ni2+-treated cells were resistant to disassembly induced by nocodazole, and at least partially resistant to cold temperature (0 degrees Celsius), which also depolymerizes MTs. Since acetylated alpha tubulin serves as a marker for the presence of stable MTs, the marked enhancement of alpha tubulin acetylation in Ni2+-treated cells indicates that stabilization of MTs may be an important mechanism by which Ni2+ induces cell injury since stabilized MTs in turn should favor MT bundling. The acetylated microtubules have been shown to be more drug resistant than nonacetylated microtubules. This increased stability is most likely due to the acetylation process. Applications: Suitable for use in Western Blot and Immunofluorescence. Other applications have not been tested. Recommended Dilutions: Western Blot (Human, mouse, rat): 1-3ug/ml Immunofluorescence (Human, mouse): 1-3ug/ml Optimal dilutions to be determined by the researcher. Source: Ascites Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
