Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. Applications: Suitable for use in Neutralization, ELISA and Western Blot. Other applications have not been tested. Recommended Dilutions: Neutralization: 0.012-0.014ug/ml to yield one-half maximal inhibition [ND50] of the biological activity of hTNF-a (0.25ng/ml). ELISA (Sandwich): 0.5-2ug/ml using 100ul/well antibody. The detection limit is 0.2-0.4ng/well of recombinant hTNF-a when used along with T9160-17B. Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant hTNF-a is 1.5-3ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Reinheit: Purified by affinity chromatography.
