MaxLight(TM)650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor(TM)647, DyLight(TM)649, Cy5(TM) and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm), Emission (676nm), Extinction Coefficient 250,000. Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Applications: Suitable for use in FLISA, Western Blot and Neutralization. Other applications have not been tested. Recommended Dilution: FLISA: 0.5ug/ml (100ul/well antibody solution). Detection of 0.2-0.4ng mTNFa/well. Western Blot: 0.10.2ug/ml. Detection limit is 1.5-3ng mTNFa/lane, under either reducing or non-reducing conditions. Neutralization: To yield one half maximal inhibition (ND50) of the biological activity of mTNF-alpha (0.25ng/ml), a concentration of 0.05-0.08ug/ml of this antibody is required. Optimal dilutions to be determined by the researcher. Reconstitution: Reconstitute in sterile water to a concentration of 0.2mg/ml. Storage and Stability: May be stored at 4C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol, aliquot and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
