Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Applications: Suitable for use in FLISA, Western Blot and Neutralization. Other applications have not been tested. Recommended Dilution: FLISA: 0.5ug/ml (100ul/well antibody solution). Detection of 0.2-0.4ng mTNFa/well. Western Blot: 0.10.2ug/ml. Detection limit is 1.5-3ng mTNFa/lane, under either reducing or non-reducing conditions. Neutralization: To yield one half maximal inhibition (ND50) of the biological activity of mTNF-alpha (0.25ng/ml), a concentration of 0.05-0.08ug/ml of this antibody is required. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
