Tyrosine hydroxylase catalyzes the rate limiting step in the synthesis of dopamine and other catecholamines. The enzymes activity is regulated at the levels of transcription, translation and post-translational modification. Tyrosine hydroxylase exists in several different isoforms ranging from 50kD to about 62kD in human. The N-terminal regulatory domain contains three phospho-serine sites: Ser19, Ser31 and Ser40. Phosphorylation at Ser40 by PKA positively regulates the enzyme catalytic activity of the enzyme. Applications: Suitable for use in Immunofluorescence, Immunocytochemistry and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4C with gentle shaking, overnight. Immunofluorescence (IF-IC): 1:400 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
