Partial recombinant sequence from the C-terminal region of human TWIST1.
Basic helix-loop-helix (bHLH) transcription factors have been implicated in cell lineage determination and differentiation. The protein encoded by this gene is a bHLH transcription factor and shares similarity with another bHLH transcription factor, Dermo1. The strongest expression of this mRNA is in placental tissue, in adults, mesodermally derived tissues express this mRNA preferentially. Mutations in this gene have been found in patients with Saethre-Chotzen syndrome. Applications: Suitable for use in ELISA, Western Blot, Dot Blot, Flow Cytometry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.2-2ug/ml Immunocytochemistry: 2-100ug/ml. HeLa cells were fixed with 4% paraformaldehyde and permeabilized with 0.1% Triton X-100 in PBS. The cells were immunostained with T9300-02. Flow Cytometry: 0.5-2ug. HeLa cells were fixed in 2% paraformaldehyde/PBS and then permeabilized in 90% methanol. Cells were stained with T9300-02 or isotype control followed by Alexa Fluor 488- conjugated goat anti-mouse IgG. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
