Protein ubiquitination and deubiquitination is a reversible process catalyzed by ubiquitinating enzymes (UBEs) and deubiquitinating enzymes (DUBs) (1,2). DUBs are categorized into 5 subfamilies USP, UCH, OTU, MJD and JAMM. UCHL1 and UCHL3 belong to the UCH family of DUBs. UCHL1 is abundantly expressed in neuronal tissues and testes, while UCHL3 is more widely distributed (3,4). UCHL1 binds monoubiquitin and UCHL3 shows affinity to both ubiquitin and NEDD8, a ubiquitin-like molecule (5,6). Both enzymes play a role in the regulation of neuronal development and spermatogenesis (4,7,8) and have been implicated in neurodegenerative diseases (3-5). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% nonfat dry milk, 1X TBS, 0.1% Tween-20 at 4C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
