Asymmetric unit membrane (AUM) preparation from bovine urinary bladder.
The binding region of Uroplakin III has been localized on the extracellular part of the antigen. Together with the uroplakins UP Ia, UP Ib and UP II, uroplakin III contributes in constituting the asymmetrical unit membrane of the plaques of urothelial superficial (umbrella) cells. Uroplakin is a membrane glycoprotein (47kD) and has been shown to be a specific marker of terminal urothelial differentiation (Wu et al., 1993, 1994). About 60% of human transitional cell carcinomas (including metastases) maintain focal (sometimes very limited) expression of uroplakin III. Until now, no uroplakin staining has been found in any non-urothelial carcinoma (Moll et al., 1995). Uroplakin III may thus serve as a specific urothelial differentiation marker in cases of metastatic carcinomas with unclear primary tumor. Applications: Suitable for use in Western Blot and Immunohistochemistry. Not suitable for frozen sections. Other applications not tested. Recommended Dilutions: Immunohistochemistry: 1:10 on paraffin-embedded tissue using a heat-induced retrieval method (e.g. microwave treatment.). Incubation Time: 1 hr at RT. Optimal dilution determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
