Immunogen: Recombinant protein corresponding to Ser44-Lys153 of human UTF1 expressed in E. coli. Species Sequence Homology: mouse.
UTF1 (undifferentiated embryonic cell transcription factor 1) is a 47-57kD transcriptional coactivator. It is expressed mainly in embryonic stem cells, is regulated by Oct-3/4, and promotes rapid cell proliferation. Human UTF1 is a 341aa phosphoprotein. It is rich in Proline and Arginine residues and contains a C-terminal Leu-zipper (aa286-313) that is known to interact with the metal-binding motif of ATF-2. Unlike mouse UTF1 which contains an alternate start site at Met 43, human UTF1 lacks this Methionine and shows only one isoform. Over aa44-153, human UTF1 shares 81% aa sequence identity with mouse UTF1. Applications: Suitable for use in Western Blot, Immunocytochemistry and ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.1ug/ml using Recombinant Human UTF1. Immunocytochemistry: 5-15ug/ml using Immersion fixed D3 mouse embryonic stem cell line and human BG0IV stem cells. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
