| Vascular Adhesion Protein 1 (VAP-1), also called AOC3 (amine oxidase copper-containing 3) or SSAO (semicarbazide-sensitive amine oxidase) is a type II transmembrane sialoglycoprotein that is upregulated on endothelial cells by inflammation. The 170kD disulfide-linked homodimer of 763 aa units is an ectoenzyme that deaminates amines, forming aldehydes and hydrogen peroxide. It also mediates lymphocyte rolling, adhesion and possibly extravasation during inflammation. Adipocytes can release a soluble form via metalloproteinase cleavage. Human and mouse VAP-1 share 83% aa identity. Applications: Suitable for use in Western Blot and Direct ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml using recombinant human VAP-1/AOC3 under reducing conditions only Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
