MaxLight(TM)490 is a new Blue-Green photostable dye conjugate comparable to DyLight(TM)488, Alexa Fluor(TM)488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm), Emission (515nm), Extinction Coefficient 73,000. VEGF (vascular endothelial growth factor) is a homodimeric, disulfide-linked glycoprotein involved in angiogenesis which promotes tumor progression and metastasis. It exhibits potent mitogenic and permeability inducing properties specific for the vascular endothelium. Multiple isoforms of VEGF containing 206-, 189-, 165-, and 121-amino acid residues have been described which arise from alternative splicing of a single gene. The smaller two isoforms, VEGF165 and VEGF121, are secreted proteins and act as diffusible agents, whereas the larger two remain cell associated. VEGF/VPF plays an important role in angiogenesis, which promotes tumor progression and metastasis. Applications: Suitable for use in FLISA and Western Blot. Other applications not tested. Recommended Dilution: Direct FLISA: 0.5ug/ml using 100ul/well antibody solution. Allows the detection of 0.2-0.4ng/well of recombinant hVEGF. Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant hVEGF is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Note: Applications are based on unconjugated antibody.
