Receptors for VEGF, VEGFB and PGF have a tyrosine-protein kinase activity. The VEGF-kinase ligand/receptor signaling system plays a key role in vascular development and regulation of vascular permeability. VEGF and its high-affinity binding receptors, the tyrosine kinases FLK1 and FLT1, are thought to be important for the development of embryonic vasculature. It has been shown that an alternately spliced form of FLT1 produces a soluble protein, termed sFLT1, which binds vascular endothelial growth factor with high affinity, playing an inhibitory role in angiogenesis. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence/Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:100-1:2000 detects a band at ~180-230kD. The theoretical MW of mouse VEGFR1 is ~150kD and human VEGFR1 is ~151kD. The difference in MW is likely due to glycosylation or other post translational modifications. With CSF-1R chimera transfected lysates a doublet is seen at ~150kD representing VEGFR-2. An unknown band is also seen at ~50kD. Immunohistochemistry (Paraffin, frozen): 1:250-1:500 Paraffin requires HIER using sodium citrate buffer pH6.0 prior to staining. Immunofluorescence(IC): 1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
