Immunogen: Synthetic peptide derived from the N-terminal region of the mouse vascular endothelial growth factor receptor-2 (VEGFR-2, FLK-1, KDR) protein.
Vascular endothelial growth factor receptor-2 (VEGFR-2), also known as fetal liver kinase 1 (FLK-1) in mouse and KDR in human, is a transmembrane tyrosine kinase that plays an important role in the development of the vascular system and the vascularization of a wide variety of tumors. Binding of VEGF to VEGFR-2 leads to tyrosine phosphorylation of the dimerized receptor and subsequent phosphorylation of SH2-containing intracellular signaling proteins, including phospholipase C, Src family tyrosine kinase and phosphatidylinositol 3-kinase (PI3K) adaptor molecules, SHC, NCK, and Ras GTPase-activating protein. VEGFR-2 is expressed in cultured human umbilical vein endothelial cells (HUVEC), Hela cells, WM 35 melanoma cells, WM 9 melanoma cells, as well as mouse NIH3T3, and Balb/c 3T3 cells. In a single study of 115 paraffin-embedded cancer specimens analyzed by immunohistochemistry, VEGFR-2 expression was observed in bladder tumors, breast cancer, intestinal cancer and lung cancer, suggesting a direct role for VEGF as an autocrine growth factor in a variety of human cancers. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 0.1-1ug/ml Western Blot: 1-5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
Reinheit: Purified by immunoaffinity chromatography.
Formulierung: Supplied as a liquid in PBS, pH 7.2. No preservatives added. Labeled with horseradish peroxidase (HRP).
