| Applications: Suitable for use in ELISA, Western Blot, Immunocytochemistry, Flow Cytometry and Immunoprecipitation. Other applications not tested. Recommended Dilutions: ELISA: 0.05ug/ml, as reported by an independent laboratory. Western Blot: 0.5-2ug/ml detected phosphorylated Vasodilator Stimulated Phosphoprotein in RIPA lysates from CTLL cells. CTLL cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with V2118 (0.5ug/ml). Proteins were visualized using a Gt xMo IgG (HRP)* and a chemiluminescence detection system. Immunocytochemistry: 3ug/ml showed positive immunostaining in SNP or 8pCPT-cGMP-treated human platelets, fixed with 4% paraformaldehyde and permeabilized with 0.2% Triton-X, reported by an independent laboratory. Flow Cytometry: 2.4ug/ml using fixed and permeabilized human platelets, as reported by an independent laboratory. Immunoprecipitation: Immunoprecipitates phosphorylated Vasodilator Stimulated Phosphoprotein from vanadate-treated A431 cells, as reported by an independent laboratory. Optimal dilutions to be determined by the researcher. *Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-14C: IgG, Fc (HRP) Pab Gt xMo I1904-19C: IgG, F(ab)2 (HRP) Pab Gt xMo Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
